Study type: Medical/biological study (experimental study)

Static and ELF magnetic fields induce tumor growth inhibition and apoptosis med./bio.

Published in: Bioelectromagnetics 2001; 22 (6): 419-428

Aim of study (acc. to author)

To investigate the effects of different static and extremely low frequency magnetic fields (MF) on tumor cell death both in vitro and in vivo.

Background/further details

The findings of two different experimental setups were listed in this paper: 1) An in vitro study using two transformed cell lines (WiDR, a human colon adenocarcinoma, and MCF-7, a human breast adenocarcinoma) and one non-transformed cell line (MRC-5 embryonal lung fibroblast). 2) An in vivo study using CD-1 mice bearing WiDr tumors.

Endpoint

Exposure

Exposure Parameters
Exposure 1: 0–100 Hz
Exposure duration: 20 min
Exposure 2: 0–100 Hz
Exposure duration: 70 min/day, 5 days/week, for 4 weeks

General information

All the magnetic fields are the effective values except for DC MF.

Exposure 1

Main characteristics
Frequency 0–100 Hz
Type
Waveform
Exposure duration 20 min
Additional info 16 Hz, 50 Hz and 100 Hz magnetic field + static magnetic field were applied.
Exposure setup
Exposure source
Setup Flasks containing cell cultures were located inside the thermostated water bath box (maintained at 37°C) placed between the pair of coils. The pair of coils were mounted horizontally with their axes in the same plane and orthogonal to the ground.
Sham exposure A sham exposure was conducted.
Additional info Up to three flasks containing cell cultures and one containing only culture medium were used.
Parameters
Measurand Value Type Method Mass Remarks
magnetic flux density 1 mT - measured - 3 mT, 10 mT and 30 mT; both static and ELF MF

Exposure 2

Main characteristics
Frequency 0–100 Hz
Type
Waveform
Exposure duration 70 min/day, 5 days/week, for 4 weeks
Additional info 16 Hz, 50 Hz and 100 Hz magnetic field + static magnetic field were applied.
Exposure setup
Exposure source
Setup Each mouse was held in an individual plexiglas box (14.7 x 3.6 x 3.6 cm) placed between the two coils. The pair of coils were mounted horizontally with their axes in the same plane and orthogonal to the ground. A total of 5 mice were simultaneously exposed in the exposure unit.
Sham exposure A sham exposure was conducted.
Additional info There were four treatment groups: Treatment 1: extremely low frequency magnetic field (ELF MF) alternated with static magnetic field (SMF); Treatment 2: SMF applied on odd days and SMF+ELF MF applied on even days; Treatment 3: ELF MF + SMF alternated with SMF alone; Treatment 4: ELF MF + SMF.
Parameters
Measurand Value Type Method Mass Remarks
magnetic flux density 300 µT effective value measured - 0.6 mT, 0.8 mT, 1 mT, 1.5 mT, 2 mT, 2.5 mT and 3 mT; ELF MF
magnetic flux density 2.5 mT minimum measured - 3.8 mT, 4 mT and 5 mT; static magnetic field

Main outcome of study (acc. to author)

The in vitro experiments were performed to assess optimal MF condition for the in vivo experiment. The results of the in vitro study demonstrate that MF-exposure is capable to induce an apoptosis-like death when the intensity is higher than 1 mT. This effect does not depend upon the frequency of the magnetic field.
The in vivo experiment revealed growth inhibition in the inoculated tumor cells. No adverse effects induced by MF-exposure was found in any investigated kind of normal somatic cells.
