Study type: Medical/biological study (experimental study)

Gene expression of cytokine receptors in HL60 cells exposed to a 50 Hz magnetic field med./bio.

Published in: Bioelectromagnetics 2002; 23 (5): 339-346

Aim of study (acc. to author)

To study the effects of a 50 Hz extremely low frequency sinusoidal magnetic field on the gene expression cytokine receptors in human acute myeloid leukaemia cells in vitro.

Background/further details

The investigated cell surface cytokine receptors exert a wide range of biological and immunological functions, e.g. in the defense against pathogens.
Tetradecanoylphorbol acetate treated cells served as the positive control.

Exposure

Exposure Parameters
Exposure 1: 50 Hz
Exposure duration: continuous for 0.5 h, 1 h, 2 h, 4 h and 8 h
Exposure 2: 50 Hz
Exposure duration: continuous for 12 h, 24 h, 36 h, 48 h and 72 h

General information

The field intensities were chosen because their similarity to occupational exposure levels.

Exposure 1

Main characteristics
Frequency 50 Hz
Type
Waveform
Exposure duration continuous for 0.5 h, 1 h, 2 h, 4 h and 8 h
Exposure setup
Exposure source
Setup 3 groups of square copper coils (36 x 36 cm), spaced 18 cm apart from each other; the coils provided a very uniform magnetic field in the area of 10 x 10 x 10 cm³ where the cell cultures were located; the coils were placed in a µ-metal container. Sham and positive control cells treated with phorbol 12 myristate 13-acetate were placed in an identical incubator where the conditions were same as for the exposure groups but without magnetic field. Control and sham exposures took place for 8 h.

Main outcome of study (acc. to author)

The magnetic field exposure at 0.1 and 0.8 mT for 72 hours increased gene expression levels of TNF-Rp75 and IL-6Rα in human cells. No significant change was observed in gene expression levels of TNF-Rp55 and TGFβR1.
